Application Requirements
The identification of variety authenticity is a key link in crop variety identification and intellectual property protection, and it is necessary to ensure the specificity, consistency and stability of varieties through DUS testing to prevent "false seeds" from entering the market and harming breeding innovation. Current mainstream detection methods include SSR and SNP molecular marker technology. For example, corn varieties can be identified by 40 pairs of core primers, but their labeling density is low and the method is single, making it difficult to cope with the surging detection needs. As a third-generation marker technology, SNP is more suitable for high-throughput automated detection and significantly improves identification efficiency by virtue of its high genome density, genetic stability, and simple data statistics. In addition, with the relaxation of transgenic policies, the demand for pure heterozygous detection in transgenic breeding has increased, and this process needs to be supported by high-precision instruments and strict laboratory systems, which has become an important direction for the development of breeding technology. The iteration of molecular detection technology is driving variety identification towards higher accuracy and efficiency.

Sample Processing: High-purity nucleic acid is extracted from samples (e.g., seeds) using 16/32/96 Pro throughput fully automated nucleic acid extractor, improving the sensitivity and reliability of subsequent detection.
System Preparation: The automated PCR liquid handling system (RocStation) enables plate-based PCR reaction system preparation, or Rocgene lyophilized ready-to-use reagents can be adopted; purified nucleic acid after extraction can also be transferred in plates, significantly improving workflow efficiency.
Amplification Results: Amplification is performed using gradient PCR instrument (ArchiCycler 96); PCR products are subjected to electrophoresis to verify variety authenticity. Reaction conditions can be optimized according to the characteristics of variety-specific detection primers, ensuring high specificity of results.

Scenario: A seed company in Henan Province carrying out a genotyping project


Requirements: The accuracy of detection was far lower than the actual demand, requiring repeated retesting, which increased workload and significantly raised detection costs. There was an urgent need to improve the accuracy of genetic analysis and enhance the cost-effectiveness of detection.
